Accurate cell proliferation detection method is based on the incorporation of nucleoside analogues into newly synthesized DNA and measurement, of which bromodeoxyuridine (BrdU) is a commonly used analogue. BrdU-labeled DNA is quantified using anti-BrdU antibodies after denaturation of DNA by harsh treatments (HCl, heat, or enzymes) and exposure to BrdU molecules. This step is time-consuming and difficult to perform consistently. Harsh handling can also adversely affect sample integrity and quality, making co-staining with other antibodies challenging.
Excellent Proliferation Methodology The Click-iT EdU Cell Proliferation Imaging Kit provides a cell proliferation assay superior to the BrdU assay. EdU (5-ethyl-2'-deoxyuridine) is a nucleoside analog of deoxythymine that is incorporated into DNA during active DNA synthesis. With Click-iT™ EdU, gentle fixation and detergent permeabilization are sufficient to allow small molecule-based Click-iT™ EdU detection reagents to gain access to the pathways for incorporation into DNA. As a result, the Click-iT™ EdU Imaging Kit is not only easy to use, but also more accurate and compatible with cell cycle analysis and other intracellular or extracellular target components for satisfying and enriched detection results.
