NotI restriction endonuclease can recognize GC^GGCCGC site, in Tango buffer system (+DTT) at 37° C achieves the best cleavage effect (isocleavage enzyme: FauNDI). Thermo Scientific legacy restriction enzyme products include a broad range of high-quality enzymes optimized for digestion in one of five buffer systems, in addition to the universal Tango buffer provided with the enzymes liquid for double enzyme digestion. All endonucleases are 100% active under recommended buffers and reaction conditions. To ensure stable performance of the endonuclease, Thermo Scientific restriction endonuclease reaction buffers are also premixed with BSA to enhance enzyme stability and bind contaminants that may be present in DNA preparation products.

Note: Use BseLI to digest pTZ19RJL2 plasmid DNA for analysis. Compared with linear DNA, supercoiled plasmids may require 5 times more NotI enzyme to achieve complete digestion.
