Stable
Unlike products sold as solutions, LIVE/DEAD™ Fixable Light Green Stain comes in convenient packaging of 40 test bottles to ensure long-lasting dye Maintain stability and performance over time. Amine-reactive dyes in solution lose their effectiveness over a short period of time, so it is recommended that the vial be used up after rehydration. If this is not possible, aliquot the vial contents into small volumes and store at -80°C, avoiding freeze-thaw cycles.

Robust
Dead cell identification stains can lose sensitivity after treatment with fixatives such as formaldehyde or ethanol-based fixation methods required for intracellular phosphorylation studies. LIVE/DEAD™ Fixable Light Green Stain is an amine-reactive dye that covalently binds intracellular and extracellular amines, retaining the staining pattern after formaldehyde fixation.

Low Compensation
LIVE/DEAD™ Fixable Light Green Stain was selected for its fluorescent properties to minimize interaction with other violet dyes and the excitation 488 nm blue laser Compensation between dyes. The light green fluorescent reactive dye has a maximum excitation wavelength of approximately ∼375 nm but can be well excited by a 405 nm violet laser. Its maximum emission wavelength is ∼512 nm, allowing acquisition in the second channel of most violet laser flow cytometers.

How it works
For cells with damaged cell membranes, the dye can interact with free amines inside the cells and on the cell surface to achieve strong fluorescent staining. In living cells, the reactivity of the dye is limited to the amines on the cell surface, resulting in less fluorescence intensity. The intensity difference between live and dead cells is typically greater than 50-fold and can be easily distinguished.
